Dr. Krane has devoted his career to studies of bone biology and rheumatic and metabolic bone diseases. His studies have included investigation of the effects of PTH and calcitonin, collagenase and its mechanisms, collagen synthesis and degradation in genetic and acquired diseases and inflammatory mechanisms in joint diseases. Using targeted mutations of the type I collagen substrate and MMP-13, a major collagenase, in mice his laboratory demonstrated the importance of collagenase cleaveage in controlling collagen deposition in skin, in mediating effects of hormones such as parathyroid hormone on the skeleton and in modulating events at the growth plate. Ongoing studies include the role of collagen degradation in cell differentiation, migration and survival with an emphasis on bone cells.
